Metabolomics, the study of metabolite profiles in biological samples, offers a
broad range of applications ranging from biomarker discovery to elucidating the
fundamental biochemistry associated with disease states. The focus of this
presentation will be to highlight the major technological advances and mass
spectrometry-based analytical methods that have allowed for the rapid
emergence of this new “omics” field. Metabolite extraction, chromatographic
separation, the use of different mass spectrometric instruments, and
bioinformatic analysis are critical steps in the metabolomics workflow, and
strategies at each stage used to facilitate metabolite identification and
biochemical interpretation of metabolomics data will be reviewed. The
methodological overview will be accompanied by brief biological examples
including the investigation of stem cell differentiation, the pathogenesis of
multiple sclerosis, and the identification of a new endogenous metabolite in a
thermophile microorganism.
